Allopurinol is the main ingredient of Allopsel 300 that works to reduce uric acid levels in blood and urine mainly by competitive inhibition of uric acid synthesis. Thus, reducing kidney stones and kidney pain.
Mechanism of action of Allopurinol: is an inhibitor of xanthinoxidase enzyme (an enzyme that converts hypoxanthine and xanthin precursors into uric acid) thereby reducing blood uric acid levels.
Pharmacodynamics of Allopurinol Allopsel is a drug for the treatment of hyperuricemia and gout. It is a xanthin-oxidase enzyme inhibitor. Allopurinol and its metabolite oxypurinol reduce plasma and urinary uric acid concentrations.
Pharmacokinetics of Allopurinol After oral administration, Allopsel is rapidly absorbed from the gastrointestinal tract. Bioavailability is about 67-90%. Peak plasma concentrations of allopurinol are reached after 1.5 hours and of oxypurinol 3-5 hours. Less bound to plasma proteins. About 20% of allopurinol is eliminated in the faeces. The half-life is about 1-2 hours. Less than 10% of oxypurinol is excreted unchanged in the urine but has a prolonged half-life due to tubular reabsorption. In which cases is Allopsel indicated? Allopsel 300 is indicated for the treatment of the following conditions:
Treatment of chronic gout, hyperuricemia secondary to radiotherapy or chemotherapy in leukocytosis and cancer. Also used in the treatment of kidney stones, psoriasis.
Allopsel 300 may have some other effects that are not listed on the approved label, but patients should only use this medicine to treat other conditions when directed by a doctor.

3. Dosage and how to use Allopsel:


Take Allopsel after meals. Should take the drug with plenty of water for the drug to be eliminated easily.
Gout and hyperuricemia: the minimum starting dose in adults is 100 mg. Average dose: 200 - 400 mg/day, divided into 2 - 4 times. Severe disease can be given from 600 mg up to a maximum of 800 mg/24 hours.
Hyperuricemia in cancer patients: dose is 600-800 mg/day, divided into 3-4 times for 2-3 days
Dose for patients with psoriasis: 100-400 mg/day, divided into 3 - 4 times.
Children 6-15 years old: 100 mg x 3 times/day
Dose for children under 6 years old: 50 mg x 3 times/day or 8mg/kg/day in 3 divided doses.
Patients should not arbitrarily change the dose of Allopsel 300 without consulting the doctor, pharmacist.
When used for children, it is necessary to strictly follow the instructions of the doctor.
In case the patient has an overdose and has dangerous symptoms, it is necessary to contact or immediately go to the nearest local medical facility. Patients or loved ones need to provide the doctor/medical staff with all prescriptions they are taking, all medications they are taking, including prescription and over-the-counter medicines.
If the patient forgets a dose while taking the medicine, take it as soon as possible (usually it can be taken 1-2 hours before the time ordered by the doctor). Patients with a history of liver disease should be prescribed liver function tests before taking Allopsel 300, during the course of treatment, especially the first few months.
Stop the drug immediately if you see signs of skin allergy: rash, itching, rash, scaling, discharge, painful urination, hematuria or swelling of lips and mouth, loss of appetite, weight loss.
Allopsel 300 does not affect the ability to drive and use machines. However, caution should still be used when using the drug in this case because it can cause unwanted effects.
Discontinue if skin rash, painful urination, blood in the urine, eye irritation or swelling of lips or mouth.
Note when using drugs for pregnant women: Using Allopsel 300 during pregnancy can cause adverse effects (miscarriage, fetal malformations, teratogenicity...) to any stage of development. development of pregnancy, especially in the first three months of pregnancy. Side effects of Allopsel 300
Uncommon: vomiting, nausea, rash. Rare: liver dysfunction, causing hepatitis. Very rare: dizziness, stomatitis, melena, increased blood pressure, bradycardia, sharp pain in the chest, decreased vision, cataracts, headache, somnolence, asthenia, hyperlipidemia, skin allergy such as rash, itching, scaling, rash, exudation, diabetes.
