Uses of Vacivir


Vacivir is a prescription medicine used in the treatment of cytomegalovirus retinitis and the prevention of cytomegalovirus infections. So what is Vacivir and how to use it?

1. What is Vacivir?


Vacivir is a systemic antiviral drug. The drug is prepared in the form of film-coated tablets, with the main ingredient in each tablet being Vacivir hydrochloride equivalent to 450mg of Valganciclovir.
Valganciclovir is a prodrug of ganciclovir, after entering the body, this active ingredient is quickly converted to ganciclovir by the action of esterase enzymes present in the liver and intestines.
Ganciclovir has the effect of inhibiting the replication of the herpes virus. Human viruses that are susceptible to this active ingredient include: cytoplasmic virus (CMV), herpes simplex virus 1 and 2, human herpes virus 6, 7, and 8, Epstein-Barr virus, hepatitis B virus and Varicella zoster virus .
Ganciclovir has antiviral activity through inhibition of viral DNA synthesis through the following 2 mechanisms:
Competitive inhibition of incorporation of deoxyguanosin-triphosphate into DNA by viral polymerase enzyme. The incorporation of ganciclovir triphosphate into viral DNA causes chain termination or greatly limits the ability of the viral DNA to elongate. Vacivir is indicated in the following cases:
Retinitis caused by cytomegalovirus in patients with acquired immunodeficiency syndrome (AIDS). Prevention of diseases caused by cytomegalovirus infection in organ transplant patients at risk of infection with this virus. Vacivir is contraindicated in the following cases:
People with hypersensitivity to Valganciclovir, Ganciclovir or to any of the ingredients. People who are sensitive to other antiviral drugs such as Aciclovir, Valaciclovir. Women are breastfeeding.

2. Dosage and how to use Vacivir


Vacivir is taken orally, should be taken with food.
Precautions need to be taken before using or handling the drug:
Do not crush or break the tablet. Valganciclovir in Vacivir is teratogenic and carcinogenic in humans, so care should be taken when handling broken tablets. Avoid direct contact with broken or crushed Vacivir tablets. If Vacivir powder gets into your skin or eyes, you need to rinse the area thoroughly with clean water. The dose of Vacivir drug should be strictly adhered to as prescribed by the doctor. The recommended drug dose for specific cases is as follows:
Retinitis caused by cytomegalovirus in adults: Initial treatment with active Activity: Use a dose of 900mg/time x 2 times/day, continuously for 21 days. Maintenance treatment or treatment of inactive form: Use a dose of 900mg / time x 1 time / day. In cases where the patient's retinitis worsens, the initial dose of the drug can be restarted, however, the possibility of viral resistance should be assessed. Prevention of cytomegalovirus disease in organ transplant patients: Adults with kidney transplantation use a dose of 900 mg/time x 1 time/day for 10 days of transplantation and continue to use until 100 days after transplantation. Prophylactic treatment can then be continued for up to 200 days after the kidney transplant. Adults with non-renal organ transplants: Use a dose of 900mg once a day for 10 days of transplantation and continue until 100 days after transplantation. The dose for children will be calculated by the doctor based on skin area and creatinine clearance. If the calculated dose is over 900mg, the maximum dose is 900mg. Vacivir therapy should be started within 10 days of the transplant, then continued until 200 days after the kidney transplant, and 100 days after the other organ transplant. Dosage of Vacivir in patients with renal impairment requires dose adjustment according to creatinine clearance.

3. Vacivir side effects


Common side effects of Vacivir include:
Oral candidiasis. Sepsis, viremia. Cellulitis. Urinary tract infections. Severe neutropenia. Anemia. Severe thrombocytopenia. Severe leukopenia. Severe pancytopenia. Reduce appetite. Anorexia.
