The effects of Oxaliplatin are relatively broad, cytotoxic and antitumor in many types of rectal and colon tumors. The activity of Oxaliplatin was increased in combination with 5-fluorouracil.
The mechanism of action of Oxaliplatin is explained by interacting with DNA to disrupt DNA synthesis and prevent the growth of cancer cells.
Lyoxatin 100mg is indicated in the following cases:
Adjuvant treatment of stage III colorectal cancer: Oxaliplatin in combination with Leucovorin and Fluorouracil is indicated in the adjuvant treatment regimen of stage colon cancer III in patients who have undergone radical surgery; Treatment of late colorectal cancer: The first-line treatment regimen is a combination of Oxaliplatin with Leucovorin and Fluorouracil for patients who have not been treated or operated before. An alternative treatment regimen is a combination of Fluorouracil, Irinotecan and Leucovorin for patients with late colorectal cancer that has recurred or progressed within 6 months of the first therapy.

Lyoxatin is administered by intravenous infusion in adults. In case of use in combination with 5-fluororacil, Oxaplatium should be infused first, the drug should be diluted before infusion. If the patient develops symptoms of extravasation of the drug, the drug should be discontinued immediately.
The recommended dose of Lyoxatin 100mg is 85mg/m2 by intravenous infusion every 2 weeks. The dose depends on the tolerability of the patient, so the dosage regimen for each patient may be different.
Lyoxatin is usually infused in 2 - 6 hours, the drug solution is diluted in 250 - 500ml glucose 5%, using the combination of Oxaplatium and 5 - Fluororacil to increase the effectiveness of treatment

Patients are allergic to Oxaliplatin or any component of Lyoxatin; Women who are breastfeeding; Patients with myelosuppression before starting the first course of treatment have a neutrophil count less than 2 x 109/l and/or a platelet count less than 100 x 109/l; Patients with peripheral sensory neuropathy with impaired nerve function before the first course of treatment; Patients with severe renal impairment (creatinine clearance <30ml/min). Precautions when using drugs:
Lyoxatin drug is only used in the specialty of cancer treatment and the treatment process needs to be monitored by a specialist. For patients with renal impairment: There are no specific data on the safety of Oxaliplatin when treating patients with moderate renal impairment, so the use of the drug should be weighed against the benefit / risk for the patient. In case of manifestations of anaphylaxis, the infusion should be stopped immediately and symptomatic treatment should be initiated as soon as possible. Patients treated with Lyoxatin if there are unexplained respiratory symptoms such as dyspnea, dry cough, crackles in the lungs or infiltrates on chest X-ray should stop Lyoxatin treatment until in-depth examination. rule out interstitial lung disease. In cases where abnormal liver function test results or paroxysmal hypertension are not known to be due to liver metastases, the patient should be considered for risk of severe hepatic vascular disorders.

5. Drug interactions

Oxaliplatin does not affect the risk level of 5-fluorouracil and does not compete for protein binding with some drugs such as Salicylates, Erythromycin, Sodium Valproate.
Use of live attenuated vaccines in patients immunocompromised by chemotherapeutic agents such as Oxaliplatin can lead to serious infection, even death. Therefore, vaccination with live vaccines should be avoided in patients receiving Oxaliplatin. Dead or inactivated vaccines may be given, but the response to these vaccines may be impaired.
