Uses of Asfotase alfa


Asfotase alfa is an enzyme replacement medicine used to treat hypophosphataemia, which causes soft or brittle bones that affect growth and tooth loss.

1. What is asfotase alfa?


Asfotase Alfa is an enzyme replacement therapy for the treatment of hypophosphatase (HPP), alkaline hypodosphatasia (ALP), which is a rare genetic disorder. Asfotase Alfa replaces the missing ALP, improves enzyme substrate levels and improves the body's ability to mineralize bones, thereby preventing serious diseases of the skeletal system.
Patients with perinatal/infant and adolescent HPP treated with Asfotase alfa had decreased plasma TNS ALP (tissue nonspecific alkaline phosphatase), PPi and pyridoxal 5'-phosphate (PLP) for 6-12 weeks of treatment. Asfotase alfa-treated patients demonstrated decreased bone volume and thickness indicating improved bone mineralization.

2. Indications for Asfotase alfa


Indicated for the treatment of patients with perinatal/infant and adolescent hypokalemia with hypophosphatase (HPP) syndrome.

3. Contraindications Asfotase alfa


Contraindicated in people who are hypersensitive to Asfotase alfa or any of its ingredients.

4. Usage and dosage


Dosage form: Solution for subcutaneous injection (18mg/ 0.45 mL; 28mg/ 0.7mL; 40mg/mL; 80mg/ 0.8 mL)
How to use:
Asfotase alfa is usually injected 3-6 times/week . Asfotase alfa is injected under the skin as prescribed by your doctor. Dosage:
Usual Adult Dose: 2 mg/kg subcutaneously 3 times weekly OR 1 mg/kg subcutaneously 6 times weekly. Maximum dose: 9 mg/kg/week. Usual Pediatric Dose: 2 mg/kg subcutaneously 3 times weekly OR 1 mg/kg subcutaneously 6 times weekly. Maximum dose: 9 mg/kg/week. Note:
For patients with perinatal/infant-onset dyspepsia (HPP), increase to 3 mg/kg taken 3 times/week.

5. Side effects of drugs


During the use of Asfotase alfa, patients may experience side effects such as:
Pain, itching, swelling, redness, bruising at the injection site; Change of vision; Painful or difficult urination; Pink, red, or brown urine; Severe pain in the side or lower back.
